چکیده

Vacuum.-By creating a vacuum the process of decalcification can be characterized as follows: Insoluble calciurn-salts+acid o-soluble calciumsalts+CO2. The carbon dioxide, rapidly extracted, will disturb the balance of the chemical equilibrium, and, according to Le Chatelier, result in an acceleration of the reaction. Waerhaug (1949), however, points out that there will be a more intensive contact between the specimen and the surrounding fluid, the CO2 bubbles being rapidly removed. Waerhaug (1949), Frank and Deluzarche (1950), and Engelbreth-Holm and Plum (1951) also reported a considerable shortening of the time of decalcification. Molenaar (1957) found the decalcification process only slightly accelerated.

Postembedding Decalcification of Mineralized Tissue Sections Preserves the Integrity of Implanted Biomaterials and Minimizes Number of Experimental Animals

Bone histology of decalcified or undecalcified samples depends on the investigation. However, in research each method provides different information to answer the scientific question. Decalcification is the first step after sample fixation and governs what analysis is later feasible on the sections. Besides, decalcification is favored for immunostaining and in situ hybridization. Otherwise, sam...

Decalcification by perfusion. A new method for rapid softening of temporal bones.

We describe a new technique, decalcification by perfusion, for the softening of bony tissue. The blood circulatory system was perfused in 16 rats via a cannula through the left heart ventricle with a fixative followed by New Decalc (an acidic demineralizer) for 30-240 minutes. Perfusion decalcification for 120 minutes softened all heads and middle ear specimens could be easily sampled and prepa...

Fixation and decalcification of adult zebrafish for histological, immunocytochemical, and genotypic analysis.

To facilitate the molecular analysis of tissues in adult zebrafish, we tested eight different fixation and decalcification conditions for the ability to yield DNA suitable for PCR and tissue immunoreactivity, following paraffin embedding and sectioning. Although all conditions resulted in good tissue histology and immunocytochemistry, only two conditions left the DNA intact as seen by PCR. The ...

Maintaining mRNA Integrity during Decalcification of Mineralized Tissues

Biomineralization of the extracellular matrix occurs inappropriately in numerous pathological conditions such as cancer and vascular disease, but during normal mammalian development calcification is restricted to the formation of the skeleton and dentition. The comprehensive study of gene expression in mineralized skeletal tissues has been compromized by the traditional decalcification/fixation...

The application of backscattered ultrasound and photoacoustic signals for assessment of bone collagen and mineral contents.

BACKGROUND This study examines the backscattered ultrasound (US) and back-propagating photoacoustic (PA) signals from trabecular bones, and their variations with reduction in bone minerals and collagen content. While the collagen status is directly related to the strength of the bone, diagnosis of its condition using US remains a challenge. METHODS For both PA and US methods, coded-excitation...
